Manual Analysis for the Modified DNA Comet Assay to Detect Frozen Chicken Meat Irradiated with ^(60)Co-¥ã-ray

Abstract
Modification of DNA comet assay was focused on to develop an appropriate method for the identification of frozen chicken meat irradiated at high dose and the rough dose estimation using manual analysis. Sixty frozen chicken wings were irradiated with ^60Co-¥ã-ray at a dose rate of 372 Gy/min (1.0, 3.0 and 5.0 kGy). Except for the results of preliminary experiment, analytical results from 40 samples applied with the modified DNA comet assay are reported. We completely succeeded in identifying the irradiation status of tested samples. Rough dose estimation for the irradiated sample was also 95% successful. The analytical results from the ratio of comet tail to head were statistically more reliable (R^2 = 0.82, P<0.001) than those of comet lengths. Results revealed that manual analysis for the modified DNA comet assay can be a useful, economical screening method to detect irradiated chicken meat. Although comet patterns of frozen chicken meat kept well-frozen at -20¡É showed almost no changes for 3 months, the suspected samples should be analyzed by other validated methods after the screening test, because other factors may cause DNA fragmentation of cells.
